1.定期觀察細(xì)胞培養(yǎng)過程中集落的形成。

2.顯微鏡下計(jì)數(shù)大于50個(gè)細(xì)胞克隆數(shù)，然后按下式計(jì)算集落形成率：

集落形成率（%）=（集落數(shù)/接種細(xì)胞數(shù)）×100

大鼠IL10/Interleukin-10蛋白注意事項(xiàng)

1.瓊脂對(duì)熱和酸不穩(wěn)定，如果反復(fù)加熱，容易降解，產(chǎn)生毒性，同時(shí)瓊脂硬度下降。故瓊脂高壓滅菌( 10磅 15分鐘)后按一次用量進(jìn)行分裝。

2.細(xì)胞懸液中，細(xì)胞分散度>95%。

3.軟瓊脂培養(yǎng)時(shí)，注意瓊脂與細(xì)胞混合時(shí)溫度不要超過 40℃ ，以免燙傷細(xì)胞。

4.接種細(xì)胞密度不宜過高。

5.大鼠IL10/Interleukin-10蛋白在低密度條件下培養(yǎng)，生存率明顯下降，無(wú)限細(xì)胞系和腫瘤細(xì)胞株克隆形成率一般在10%以上。但初代培養(yǎng)細(xì)胞和有限細(xì)胞系僅為0.5~5%,甚至為零。為提高集落形成率，必要時(shí)在培養(yǎng)基中添加胰島素、*等促細(xì)胞克隆形成物質(zhì)。
